The Nuclease Activities of Cathepsin Preparations from Calf Spleen and Thymus

چکیده
In a study of the role of cathepsins in nuclear metabolism (1) these intracellular proteinases were found to hydrolyze thymus nucleoproteins and render dialyzable the desoxyribonucleic acid. Further characterization of this nuclease activity of the cathepsin preparations revealed pH optima in the acid range for both desoxyribonucleic acid (DNA) and ribonucleic acid (RNA). This was in contrast to the specific activities of the pancreatic nucleases which show pH optima above neutrality (24). Evidence which establishes an intracellular desoxyribonuclease and ribonuclease activity distinct from that of the pancreas and a correlation of the factors which affect the proteolytic and nuclease activities of the cathepsin preparations are reported here.

متن کاملStudies on Some Physicochemical Properties of a Thymus Humoral Factor Conferring Immunocompetence on Lymphoid Cells
By means of an assay of graft-versus-host activity some properties of the thymic humoral factor which confers immunocompetence upon lymphoid cells in vitro have been studied. Allogeneic and xenogeneic thymic preparations were found to activate lymphoid cells from neonatally thymectomized mice, enabling initiation of a graft-versus-host response. Thus, this thymus factor is apparently neither st...

Antibodies to purified calf thymus Fl, and to chromatographically purified subfractions of calf thymus Fl histones were induced by immunization of rabbits with histone-RNA complexes. Complement fixation studies with these antibodies demonstrated structural differences between the subfractions in one organ and between those of different organs and species. The anti-F1 sera showed varying reactiv...
متن کاملThe hydrolysis of nucleoproteins by cathepsins from calf thymus.
A combination of enzymatic and staining cytological techniques with ultraviolet spectroscopy has suggested the possible participation of nucleoproteins in the synthetic processes of cells (l-4). Little is known concerning the function of enzymes in the intracellular metabolism of nucleoproteins. One approach to the problem has been through the study of the action of enzymes upon nucleoproteins....
متن کاملThe calf 5'- to 3'-exonuclease is also an endonuclease with both activities dependent on primers annealed upstream of the point of cleavage.
The catalytic activity of the calf thymus 5'- to 3'-exonuclease was measured on substrates consisting of two primers annealed adjacent to each other on a template. Exonucleolytic degradation of the downstream primer is very slow if the primers are separated by a gap of one nucleotide or if no upstream primer is present. When only a nick separates the primers, degradation is rapid. This suggests...
